A mitjans del anys 90 es va descobrir una drecera en el cicle del nitrogen que permet transformar l\u2019amoni i el nitrit en nitrogen gas sense necessitat d\u2019oxigen ni de mat\u00e8ria org\u00e0nica. Aquesta nova via, anomenada oxidaci\u00f3 anaer\u00f2bica de l\u2019amoni, \u00e9s portada a terme per un \u00fanic tipus de bacteris (anammox). Per tal de poder utilitzar aquests bacteris de forma eficient per a l\u2019eliminaci\u00f3 de nitrogen \u00e9s necessari, per\u00f2, un pas previ on els bacteris oxidadors d\u2019amoni (AOB) converteixin nom\u00e9s una part de l\u2019amoni a nitrit. D\u2019aquesta manera es regula el nivell d\u2019ambd\u00f3s compostos i es pot aconseguir una proporci\u00f3 id\u00f2nia per als bacteris anammox. El proc\u00e9s PANAMMOX\u00ae, desenvolupat al Laboratori d\u2019Enginyeria Qu\u00edmica i Ambiental de la Universitat de Girona (LEQUIA), permet tractar exitosament aig\u00fces residuals gr\u00e0cies a una combinaci\u00f3 seq\u00fcencial dels dos processos, en compartiments separats.<\/p>\n<p>Optimitzaci\u00f3 de l\u2019obtenci\u00f3 de l\u2019in\u00f2cul anammox i identificaci\u00f3 de nous bacteris<br \/>\nEls objectius principals de la tesi doctoral \u201cFrom inocula to biological reactor: molecular characterization of N-cycle bacterial assemblages in a PANAMMOX\u00ae process\u201d d\u2019Alexandre S\u00e0nchez Melsi\u00f3 han estat, d\u2019una banda, la recerca d\u2019un m\u00e8tode que permet\u00e9s aconseguir m\u00e9s f\u00e0cilment uns in\u00f2culs adequats per al reactor anammox, ja que aquests bacteris no s\u00f3n f\u00e0cilment localitzables a la natura i el seu enriquiment \u00e9s lent (tenen un temps de duplicaci\u00f3 de 15-30 dies); i, d\u2019altra banda, la caracteritzaci\u00f3 de les poblacions dels diferents tipus de bacteris que intervenen en ambd\u00f3s compartiments del proc\u00e9s PANAMMOX\u00ae a trav\u00e9s de t\u00e8cniques moleculars.<\/p>\n<p>Per tal d\u2019obtenir l\u2019in\u00f2cul necessari per al reactor anammox i optimitzar el procediment per aconseguir-lo, es van fer enriquiments per separat de fangs i sediments provinents de m\u00faltiples localitzacions. Al final d\u2019un llarg per\u00edode d\u2019enriquiment es van poder detectar bacteris anammox, mitjan\u00e7ant t\u00e8cniques qu\u00edmiques i moleculars, en cinc d\u2019aquests enriquiments provinents de sistemes naturals (llacuna costanera), ambients modificats (sistema d\u2019aiguamolls constru\u00efts) i ambients artificials (reactors an\u00f2xics i depuradores). Les t\u00e8cniques moleculars aplicades als enriquiments van permetre desenvolupar protocols per a una detecci\u00f3 primerenca dels bacteris anammox quan es troben a baixes concentracions. D\u2019aquesta manera es van poder identificar algunes esp\u00e8cies de bacteris que serien \u00fatils com a indicadors potencials de la capacitat d\u2019una mostra per desenvolupar poblacions m\u00e9s denses de bacteris anammox.<\/p>\n<p>Les t\u00e8cniques moleculars tamb\u00e9 van permetre detectar altres bacteris que acompanyen sovint els bacteris anammox, a partir d\u2019enriquiments amb or\u00edgens diversos. Es va identificar un patr\u00f3 de grups bacterians que es repeteix en nombrosos estudis, un fet que podria indicar l\u2019exist\u00e8ncia d\u2019algun tipus de relaci\u00f3 amb els bacteris anammox a nivell estructural o metab\u00f2lic. Per \u00faltim, es va portar a terme una caracteritzaci\u00f3 molecular de les poblacions de bacteris oxidadors de compostos nitrogenats que es troben en el reactor on l\u2019amoni \u00e9s convertit parcialment a nitrit, com a pas previ a la reacci\u00f3 anammox. Es va analitzar com canviaven les poblacions d\u2019aquests bacteris durant el proc\u00e9s d\u2019adaptaci\u00f3 del reactor per poder tractar aig\u00fces residuals amb elevades concentracions de nitrogen, en qu\u00e8 es passava d\u2019unes esp\u00e8cies de Nitrosomonas pr\u00f2pies d\u2019ambients amb baixa concentraci\u00f3 de nitrogen a unes altres que es coneixien d\u2019ambients amb alta c\u00e0rrega nitrogenada. A m\u00e9s, es va poder determinar que algunes esp\u00e8cies que el reactor te\u00f2ricament hauria d\u2019eliminar (com les del g\u00e8nere Nitrospira), no nom\u00e9s es mantenien sin\u00f3 que fins i tot podien arribar a cr\u00e9ixer-hi, tot i que no generaven cap problema per al seu correcte funcionament.<\/p>\n<p>S\u2019ha aconseguit, doncs, una major comprensi\u00f3 a trav\u00e9s de t\u00e8cniques moleculars dels grups bacterians que intervenen en el proc\u00e9s PANAMMOX\u00ae desenvolupat pel LEQUIA;
